Single Drop Micro Extraction (SDME)
Single drop micro extraction (SDME) has emerged as one of the simplest and most easily implemented forms of micro-scale sample cleanup and pre-concentration. An ordinary PAL Gas Chromatography syringe is used to suspend micro liter quantities of extracting solvent either directly immersed in the sample, or in the headspace above the sample. The syringe is then used to inject the solvent and extracted analytes into the Gas Chromatography system for identification and/or quantitation.
SDME is a straightforward technique in which a micro drop of solvent is suspended from the tip of a conventional GC syringe and then is immersed in a sample solution in which it is immiscible or suspended in the headspace above the sample.
Headspace SDME is similar to traditional headspace sampling in that volatiles are sampled from the vapors above the sample, thus avoiding interferences from the sample matrix.
Headspace SDME is similar to SPME but the fiber used in SPME is replaced by a liquid micro drop.
Advantages of the technique include high selectivity, good quantitation, low detection limits, no carryover, higher upper limits of detection and minimal sample preparation. One of the biggest advantages is you only need one drop of a solvent instead of a SPME fiber. HS-SDME is found to provide excellent clean up for samples with complicated matrices.
Like with SPME, you can perform the SDME technique manually. And once you’ve worked out your method,you can automate it with your Combi PAL autosampler.
SDME, using typically 1–3 lL of an organic solvent at the tip of a microsyringe. After extraction, the microdrop is retracted back into the syringe and transferred for further analysis.
There are currently two main approaches can be used to perform SDME with the PAL: (1) Direct Immersion (DI)-SDME (2) Headspace (HS)-SDME
In DI-SDME, a drop of a water-immiscible solvent is suspended directly from the tip of a microsyringe needle immersed in the aqueous sample.
